     TNF-alpha TYPICAL DATA

    For each experiment, a standard curve needs to be set for each microplate, and the specific OD value may vary depending on different laboratories, testers, or equipment. The following example data is for reference only. The sample concentration was cal-culated based on the results of the standard curve. The minimum detectable con-centration of mouse TNF-alpha is less than 4 pg/mL.

Validation
  • Sample Values

    RAW 264.7 mouse monocyte/macrophage cells (3.5 x 10⁶ cells/mL) were cultured in DMEM supplemented with 10% fetal bovine serum, 2 mM L-glutamine, 100 U/mL penicillin, and 100 ug/mL streptomycin sulfate. Cells were cultured unstimulated or stimulated with 5.0 ug/mL LPS and 100 ng/mL rmlL-10 for 5 and 7 days. Subsequent analysis involved quantitative measurement of mouse TNF-alpha concentrations in collected culture supernatants using standardized immunoassays.

    Ten replicates of each of three samples containing different TNF-α concentrations were tested in one assay. Acceptable criteria: CV<10%.
