    ELISA binding of Monoclonal Anti-MMAE&MMAF Antibody, Mouse IgG1 (Cat. No. MME-M5252) with Trastuzumab Deruxtecan, IgG1-MMAF, Sacituzumab Govitecam, Disitamab Vedotin (RC48), Trastuzumab MMAE and Trastuzumab-DM1 conjugated antibody respectively.
    The coating antibody was Monoclonal Anti-MMAE&MMAF Antibody, Mouse IgG1 (Cat. No. MME-M5252), used at 1 μg/mL concentration. The primary antibody were different payload conjugated antibodies, including Trastuzumab Deruxtecan, IgG1-MMAF, Sacituzumab Govitecam, Disitamab Vedotin (RC48), Trastuzumab MMAE and Trastuzumab-DM1 conjugated antibodies used at 0.5 μg/mL concentration. The secondary antibody was Peroxidase AffiniPure Goat Anti-Human IgG, Fcγ fragment specific (min X Bov, Hrs, Ms Sr Prot) (Jackson, Cat. No. 109-035-098) used at 1:12000 concentration.
    Monoclonal Anti-MMAE and MMAF Antibody, Mouse IgG1 (Cat. No. MME-M5252) specifically recognizes the MMAE and MMAF payloads. In this assay, it shows binding to ADCs carrying MMAE or MMAF (Disitamab Vedotin (RC48), Trastuzumab MMAE, and a model IgG1-MMAF conjugate), and shows no cross-reactivity with ADCs carrying different payloads (Trastuzumab Deruxtecan (DXd), Sacituzumab Govitecan (SN-38), and Trastuzumab-DM1) under the tested conditions (Routinely tested).

Background

Monomethyl auristatin E (MMAE) is a synthetic derivative of dolastatin 10 and functions as a potent mitotic inhibitor by inhibiting tubulin polymerization. MMAE is widely used as a cytotoxic component of antibody-drug conjugates (ADCs) to treat several different cancer types. Anti-MMAE antibody is a Mouse monoclonal antibody. This antibody has been shown to work in applications such as: PK, PD, Immunoassay and ELISA.
