The primary antibody were different payload conjugated antibodies, including Tusamitamab Ravtansine, Trastuzumab Deruxtecan, Sacituzumab Govitecam, Disitamab Vedotin (RC48), ADC-Duocarmycin and Trastuzumab MMAE conjugated antibodies used at 0.5 μg/mL concentration. The secondary antibody was HRP conjugated Anti-Human-IgG-Fc Antibody (6F11C8), mAb (Acro, Cat. No. IGG-LY69) used at 1:10000 concentration.

Background

DM4 (also known as ravtansine or soravtansine) is a maytansinoid derivative that serves as a potent cytotoxic payload in antibody-drug conjugates (ADCs). It functions by inhibiting tubulin polymerization, arresting cell division, and inducing apoptosis in tumor cells. Structurally modified from DM1, DM4 enhances conjugation compatibility with linkers and has been used in several clinical ADC drugs, including mirvetuximab soravtansine for ovarian cancer treatment.
