Product Details

  • Source

Chimeric Monoclonal Anti-Datopotamab Antibody, Mouse IgG1 (7D3) is recombinantly expressed in HEK293. It consists of mouse variable regions genetically fused to mouse IgG1 constant domains.

  • Antibody Type

    Recombinant Monoclonal

  • Clone

    7D3

  • Isotype

    Mouse IgG1, Kappa

  • Host Species

    Mouse

  • Specificity

7D3 is an inhibitory anti-idiotype monoclonal antibody that specifically recognizes Datopotamab. No detectable binding to the tested human IgG1 controls, Bevacizumab, Cetuximab, or TROP-2 was observed under the tested conditions. 7D3 inhibits the binding of Datopotamab to TROP-2.

     Datopotamab ELISA

Datopotamab was quantified using a PK sandwich ELISA with Monoclonal Anti-Datopotamab Antibody, Mouse IgG1 (7D3) (Cat. No. DAB-MY2439a) at 1 μg/mL as the capture antibody and Biotinylated Monoclonal Anti-Datopotamab Antibody, Mouse IgG1 (5D6) (Cat. No. DAB-BLY2438a) at 0.5 μg/mL as the detection antibody. Datopotamab standards were prepared in 10% human serum. Detection was performed using HRP-conjugated streptavidin (QC tested).

    Protocol
  • Datopotamab ELISA

The specificity of Monoclonal Anti-Datopotamab Antibody, Mouse IgG1 (7D3) (Cat. No. DAB-MY2439a) was evaluated by ELISA. Datopotamab was immobilized at 1 μg/mL (100 μL/well). 7D3 specifically recognized Datopotamab, with no detectable binding to the tested human IgG1 controls, Bevacizumab, Cetuximab, or Human TROP-2 under the tested conditions (Routinely tested).

    Protocol
  • Datopotamab ELISA

The ability of Monoclonal Anti-Datopotamab Antibody, Mouse IgG1 (7D3) (Cat. No. DAB-MY2439a) to block Datopotamab to TROP-2 was evaluated by ELISA. Datopotamab was immobilized at 1 μg/mL (100 μL/well). Increasing concentrations of 7D3 were tested for their ability to inhibit the biotinylated Human TROP-2 to Datopotamab. 7D3 inhibited Datopotamab binding to TROP-2 in a dose-dependent manner, with an IC₅₀ of approximately 0.26 μg/mL under the tested conditions (Routinely tested).

    Protocol
  • Bioactivity-SPR

     Datopotamab SPR

The binding affinity of Monoclonal Anti-Datopotamab Antibody, Mouse IgG1 (7D3) (Cat. No. DAB-MY2439a) to Datopotamab was determined by surface plasmon resonance (SPR). Datopotamab was captured on an anti-mouse IgG-coupled CM5 sensor chip, and Datopotamab was injected as the analyte at serial concentrations (0.781-50 nM). The apparent equilibrium dissociation constant (KD) was determined to be 3.40E-09 M (3.4 nM) (Routinely tested).

FAQ

  • product

Can lyophilized proteins remain stable during room-temperature shipping or temporary room-temperature exposure?

In general, lyophilized proteins remain stable during room-temperature shipping or temporary exposure to room-temperature conditions.

Many ACROBiosystems recombinant proteins are supplied in lyophilized (freeze-dried) form. The lyophilization process effectively removes moisture from the product, helping reduce degradation-related reactions and improve protein stability during transportation and storage.

To evaluate the stability of lyophilized proteins under normal temperature conditions, ACROBiosystems conducted a stability study on 11 representative lyophilized protein products at 37°C. The study demonstrated that these proteins maintained good quality stability after storage at 37°C for approximately 20 days. Based on these validation results, temporary room-temperature exposure during shipping or handling is generally not expected to have a significant impact on product quality or downstream application performance.

To ensure optimal long-term stability, products should be stored according to the storage conditions specified in the product Certificate of Analysis (COA) upon receipt.

    Supporting Document

Background

A humanized monoclonal IgG1 antibody that binds to human trophoblast cell surface antigen 2 (TROP2), a transmembrane glycoprotein overexpressed on the surface of various epithelial tumor cells. Datopotamab contains human constant regions and variable regions (complementarity-determining regions) derived from a murine antibody specific for TROP2. It is produced in a Chinese Hamster Ovary (CHO) mammalian cell expression system and has a molecular weight of approximately 148 kilodaltons.
