    The coating antibody was Monoclonal Anti-DM-1 Antibody, Rabbit IgG (M5D04) (Cat. No. DM1-MY2358), used at 1 μg/mL concentration. The primary antibody were different payload conjugated antibodies, including DM1 Antibody Human IgG, Trastuzumab Deruxtecan, Sacituzumab Govitecan, Trastuzumab MMAE and IgG1-MMAF conjugated antibodies used at 0.5 μg/mL concentration.     In general, lyophilized proteins remain stable during room-temperature shipping or temporary exposure to room-temperature conditions.


    Many ACROBiosystems recombinant proteins are supplied in lyophilized (freeze-dried) form. The lyophilization process effectively removes moisture from the product, helping reduce degradation-related reactions and improve protein stability during transportation and storage.


    To evaluate the stability of lyophilized proteins under normal temperature conditions, ACROBiosystems conducted a stability study on 11 representative lyophilized protein products at 37°C. The study demonstrated that these proteins maintained good quality stability after storage at 37°C for approximately 20 days. Based on these validation results, temporary room-temperature exposure during shipping or handling is generally not expected to have a significant impact on product quality or downstream application performance.


    To ensure optimal long-term stability, products should be stored according to the storage conditions specified in the product Certificate of Analysis (COA) upon receipt.

Background

DM1, a maytansinoid derivative, is a common payload toxin in antibody-drug conjugates (ADCs). It binds to tubulin, hinders microtubule assembly, arrests cell division, and induces apoptosis, but has off-target toxicity. ADCs like T-DM1 have shown good efficacy in HER2-positive cancers. For recent progress, many new DM1-containing ADCs are under development. For example, GQ1001 has entered Phase I clinical trials for HER2+ advanced solid tumors, and some have demonstrated antitumor activity in T-DM1-resistant models in preclinical studies.
