TR-FRET (Time-Resolved Fluorescence Resonance Energy Transfer) is a detection technology that combines Fluorescence Resonance Energy Transfer (FRET) with Time-Resolved Fluorescence (TRF) measurement. It relies on the distance between donor and acceptor fluorophores—when they are brought close together, energy transfer occurs, generating a measurable signal. The time-resolved component introduces a delay in signal detection, effectively minimizing interference from autofluorescence and scattered excitation light.
By integrating the strengths of both FRET and TRF, TR-FRET offers high sensitivity, ease of use, and reliable results. It is well-suited for high-throughput screening and widely applicable in protein quantification, signaling pathway analysis, disease mechanism studies, and more—making it a valuable tool in modern drug discovery and development.
Fluorescence Resonance Energy Transfer (FRET): When molecules are not bound, the donor and acceptor remain far apart—typically more than 10 nm—so the excited donor cannot effectively transfer energy to the acceptor. As a result, only the donor emission peak (620 nm) is observed; When molecular binding occurs, the donor and acceptor are brought into close distance—usually less than 10 nm—allowing energy transfer from the excited donor to the acceptor via FRET. This results in emission from the acceptor, producing a detectable signal at 665 nm.
Time-Resolved Fluorescence (TRF) Detection: After the excitation pulse ends, fluorescence signals from the donor and acceptor are measured following a short delay—typically between 50 and 150 microseconds. This delay effectively eliminates interference from nonspecific background fluorescence. By analyzing the ratio of fluorescence intensities, TRF enables accurate assessment of molecular interactions, including binding strength, affinity, and kinetics.
